When I use the option Save My Desktop to store my new layout, this layout is not used when I re-open the channel , the default layout is used instead.

From the Help file I understood:
'The new layout is applied the next time you open the Channel'

Here are some tips for saving your Console settings:

 

If you wish to add or remove columns within the Managed detail pane, try making the changes from the Configuration Groups tab since it has the same columns by default. Then save your settings. Close and reopen the Console. The changes should affect both tabs.

 

If you are adding and removing columns, just do the add columns first, then save and close the Console. Reopen, then make the changes that involve removing columns. Save.

 

If this still isn’t saving settings, rename your .layout file located in the Prism Suite folder. (DO NOT DELETE the default.layout file.) If the channel is unsecured, the file is “unathenticated user.layout”. If you have a secured channel, the file is <username>.layout. Re-launch the Console and save your Desktop to create a new layout file. Then follow the suggestions above.

Problem is solved by saving setting in Configuration Groups
